Cell invasion is cell migration through a physiological barrier in response to a chemoattractant, and this recapitulates cell movement within a physiological environment which is composed of extracellular matrix proteins. Here, the membranes are coated with a layer of extracellular matrix proteins, and the cells must traverse this barrier through a combination of protein degradation and cellular locomotion.
Preparation of Cells for Assay:
1.Treat and grow cells according to experimental condition. It may be desirable to synchronize cells to the same growth state.
2.Trypsinizing and count of cells.
3.Remove trypsin and resuspend with fresh medium.
4.Plate 1X105 cells in 200 μl of defined medium into upper chamber.
5.Add 300 μl of medium lower chamber.
6.Culture for approximately 20 hours. The time varies for different cell lines and may be up to 48 hours for less invasive cells.
Cell Invasion Assays accelerate the screening process for compounds that influence cell invasion through in vitro extracellular matrices. These assays offer a flexible, standardized, high-throughput format for quantitating the degree to which invasive cells penetrate a barrier consisting of extracellular matrix proteins in response to chemoattractants and/or inhibiting compounds.
Cell Invasion Assay based on the ability of invasive and metastatic cancer cells, as well as endothelial cells to cross basement membranes and/or connective tissues in spread or to form new blood vessels.
Cell Invasion Assay is flexible, easy, quantitative, rapid and reliable way to quantify the invasive capacity of many cell types.
• Cell Invasion Assay is used to measure tumor cell invasion and can be successfully used to study endothelial activation and angiogenesis. Cell Invasion Assay is flexible, easy, quantitative, rapid and reliable way to quantify the invasive capacity of many cell types.
